摘要
建立了藜麦中槲皮素、山柰酚的高效液相色谱含量测定方法,藜麦样品用80%甲醇溶液超声波及回流提取后再加浓盐酸水解,采用高效液相色谱法测定水解液中槲皮素和山柰酚的含量(以十八烷基硅烷键合硅胶为填充剂,甲醇-0.4%磷酸溶液(55∶45)为流动相,检测波长360 nm,流速1.0 mL/min,柱温30℃,进样量20μL),采用外标法进行面积定量。结果表明,槲皮素在0.02~0.40μg,山柰酚在0.01~0.20μg范围内呈良好的线性关系(r=0.999);平均加样回收率分别为91.0%,89.3%,相对标准偏差分别为0.7%,4.5%(n=9)。该方法具有操作简单、重现性好、结果可靠、专属性强等特点,可用于藜麦中槲皮素和山柰酚的含量测定。
A HPLC method for the determination of quercetin and kaempferol in quinoa was established.The sample was hydrolyzed with concentrated hydrochloric acid in 80%methanol solution by ultrasound and backflow.The hydrolysate was prepared by high performance liquid chromatography with eighteen alkyl silane bonded silica gel as filling agent and methanol-0.4%phosphoric acid solution(55∶45)as mobile phase.The detection wavelength was 360 nm with 1.0 mL/min as flowrate,column temperature was 30℃with 20μL injection volume,the results were used to determine by external standard method.The result showed that the calibration curve for quercetin was linear(r=0.999)in the range of 0.02-0.40μg and the mean recovery was 91.0%,RSD was 0.7%(n=9),for kaempferol the calibration curve was linear(r=0.999)in the range of 0.01-0.20μg and the mean recovery was 89.3%,RSD was 4.5%(n=9).This method is accurate,simple,sensitive,reliable,and is applied to the determination of quercetin and kaempferol in quinoa with good result.
