Optimization of Extraction Process for Polypherol from Corn Bracts

摘要 响应面法优化玉米苞叶多酚提取工艺,在单因素试验基础上,以乙醇体积分数、提取温度、提取时间和超声功率为变量因素,多酚得率为响应值,利用Design Expert 8.0.6软件,采用Box-Behnken试验设计,研究变量及其交互作用对多酚得率的影响,建立四元二次多项式数学回归模型,并进行响应面分析、最佳条件的确定和验证试验。结果表明,玉米苞叶多酚提取最佳工艺为乙醇体积分数70%(V/V),提取温度58℃,提取时间44 min,超声功率55 Hz,在此条件下多酚得率为3.719±0.030(n=6)mg/g,与预测值相对误差为0.216%,比传统方法多酚得率提高28.7%,试验稳定性和重现性好,验证了数学模型的有效性。超声辅助响应面法对提高玉米苞叶多酚得率具有应用价值,为玉米苞叶的开发利用提供了理论依据和技术支持。 The aim of the study was to extract polypherol from corn bracts through optimizing process assisted by ultrasonic.Based on single-factor experiments,ethanol concentration,extraction temperature,extraction time and ultrasonic power were set as independent variables and the yield of polypherol was response value.Effects of each variable and their interactions on polypherol were determined using Box-Behnken method.The quadratic polynomial regression equations between polypherol and each variable were built using Design-Expert software.The results showed that the optimal conditions of polypherol from corn bracts were set as follows:ethanol concentration 70%,extraction temperature 59℃,extraction time 44 min and ultrasonic power 55 Hz,respectively.The yield of polypherol was 3.719±0.030 mg/g(n=6)under the modified conditions.The relative error was 0.216%compared to predictive value,indicating the model fitted well with experimental data.In addition,the extraction yield was increased by 28.7%as compared to the traditional extraction method.The results could provide a theoretical foundation and scientific basis for the development of natural antioxidant products of corn bracts.
