摘要
为了探究DNA条形码技术在蓟马类昆虫物种鉴定方面的可行性,以采自陕西汉中地区的管蓟马种类为研究材料,借助DNA条形码技术,对该地区多种植物上的管蓟马进行分类研究,通过对蓟马标本的无损伤提取、扩增、测定序列,共成功获得18条序列,其中以蓟马科Thripidae的2属2种为外群,管蓟马科Phlaeothripidae管蓟马亚科Phlaeothripinae的3属5种作为内群,构建了系统发育树。结果鉴定出5属7种,其中简管蓟马属包括3种,滑管蓟马属1种,器管蓟马属1种以及蓟马科蓟马属1种和带蓟马属1种。DNA条形码数据可辅助形态学进一步确定物种,区分形态上难以区分的物种。
Based on the COI gene sequence of mitochondrial DNA,the molecular research on Phlaeothripidae species from Hanzhong in Shaanxi province was studied.Through the no damage extraction,amplification and sequencing of the COI gene sequence,18 sequences have been gotten for constructing the phylogenetic tree,and five genera and seven species are identified.The work provides some useful information for the study on species diversity of Phlaeothripidae and theoretical foundation for the biological control of the thrips in Southern Shaanxi.
