摘要
为提高转移因子的生产效率,采用管式微滤+超滤装置代替透析袋的工序提取猪脾脏中的转移因子,研究分离膜工艺中猪脾脏均质液的稀释方法、冻融次数及膜清洗恢复情况.实验结果表明:脾脏均质液冻融3次,采用纯水1∶1(体积比)稀释,在过滤过程中不断添加纯水以降低操作温度并提高过滤效率.微滤和超滤操作约3 h完成,操作温度始终低于37℃,更适宜转移因子的生产,超滤透过液中多肽、核糖的含量也符合半成品的要求.改进后的工艺操作简单,成本低,生产周期短,可适用于大规模工业生产.
In order to improve transfer factor production efficiency,this paper used tube microfiltration and ultrafiltration device instead of dialysis bag to extract transfer factor from pig spleen.Dilution methods of pig spleen homogeneous solution,freezing and thawing times and membrane permeate rate recovery were studied in separation membrane process.Results showed that it is better which spleen homogeneous solution was freeze-thawed three times,and diluted 1∶1(V∶V)with pure water combined continuous pure water addition.Microfiltration and ultrafiltration operation can be completed in about 3 h.Operating temperature was always lower than 37℃which was suitable for maintenance of transfer factor activity.And the concentration of peptide and ribose in UF permeate liquid also meet requirements of semi-finished products.So because of simple operation,low cost and short production cycle,membrane process can be applied to large-scale industrial production.
