摘要
建立了一种简便、准确、灵敏测定饮品中果糖、葡萄糖、蔗糖和乳糖等水溶性糖含量的高效液相色谱-蒸发光散射(HPLC-ELSD)检测方法。使用Zorbax Carbohydrate(250 mm×4.6 mm,5μm)色谱柱分离,以乙腈∶水=77∶23(V/V)为流动相,流速为1.0 m L/min,蒸发光散射检测器(ELSD)为检测器,ELSD Spray Chamber温度为30℃,漂移管温度60℃,N2压力为0.5 Mpa。结果表明:果糖、葡萄糖、蔗糖、乳糖在10.00~2 100.00 mg/L范围内呈良好线性关系;几种糖的回收率均在73.1%~114.8%范围之内,相对标准偏差均小于1%,适用于饮品中糖含量的测定。
A simple,accurate and sensitive HPLC-ELSD method was established for the determination of water-soluble sugar in beverages such as fructose,glucose,sucrose and lactose.The separation was carried out on a Zorbax Carbohydrate(250 mm×4.6 mm,5μm)column with acetonitrile∶water=77∶23(V/V)as mobile phase at a flow rate of 1.0 mL/min with an ELSD detector and an ELSD Spray Chamber temperature of 30℃,drift tube temperature 60℃,N2 pressure 0.5 Mpa.The results showed that there was a good linear relationship between fructose,glucose,sucrose and lactose in the range of 10.00~2100.00 mg/L.The recoveries of sugars were in the range of 73.1%~114.8%and the relative standard deviations were less than 1%.The method is suitable for the determination of sugar content in beverages.
