下调MTDH基因表达对乳腺癌MCF-7细胞增殖及凋亡的影响 被引量:1

摘要 目的探讨下调MTDH基因表达对乳腺癌MCF-7细胞增殖和凋亡的影响。方法乳腺癌MCF-7细胞株常规消化并接种,按照LipofectamineTM2000说明书进行转染,转染组转染MTDH-homo-1432,对照组转染等剂量的control-FAM。转染24 h后检测转染效率。琼脂糖凝胶电泳检测MTDH抑制效果。Western blotting法检测转染前后MTDH蛋白表达。通过MTT和流式细胞术检测MTDH基因表达下调后对MCF-7细胞增殖、细胞周期和凋亡坏死的影响。结果 MTDH-siRNA的转染率达90%,转染后MTDH-siRNA-1432对MTDH表达的干扰作用最为显著,抑制效率达89%。Western blotting检测结果显示,转染后MTDH蛋白表达降低。转染组乳腺癌细胞增殖受到抑制。转染组、对照组G1期细胞比例分别为67.81%±1.07%、46.81%±0.26%(P<0.05),G2+S期细胞比例分别为29.08%±1.14%、55.55%±0.27%(P<0.05)。转染组细胞凋亡比例10.60%±0.46%,坏死比例12.90%±3.63%;对照组细胞凋亡比例4.80%±0.20%,坏死比例6.10%±0.44%,转染组细胞凋亡和坏死比例高于对照组(P均<0.05)。结论下调MTDH基因表达可抑制乳腺癌MCF-7细胞增殖,并促进细胞凋亡和坏死。
