小檗碱对胰岛素抵抗模型小鼠过氧化物酶体增殖物激活受体γ辅激活因子1α/骨骼肌葡萄糖转运蛋白4表达的影响 被引量:2

摘要 目的观察小檗碱对胰岛素抵抗模型小鼠过氧化物酶体增殖物激活受体γ辅激活因子1α(PGC-1α)和骨骼肌葡萄糖转运蛋白4(GLUT4)表达的影响,探讨小檗碱改善胰岛素抵抗的作用机制。方法建立胰岛素抵抗小鼠模型,进行小檗碱灌胃,检测血糖、胰岛素及胰岛素敏感性;RT-PCR检测骨骼肌GLUT4及PGC-1αm RNA表达水平。结果小檗碱能够显著降低血糖,提高胰岛素敏感性,促进骨骼肌GLUT4及PGC-1α表达。结论小檗碱能够通过增加PGC-1α表达,促进GLUT4表达,改善胰岛素抵抗。 Objective To observe the effect of berberine on the expression of GLUT4 and PGC-1αin mice with insulin resistance,and to explore the molecular mechanism of berberine in improving insulin resistance.Methods Mice models with insulin resistance were established,and berberine was used by intragastric administration to detect blood glucose,insulin and insulin sensitivity.RT-PCR was used to detect the expression level of GLUT4 and mRNA of PGC-1αin skeletal muscle.Results Berberine significantly reduced blood sugar,increased insulin sensitivity,and promoted the expression of GLUT4 and PGC-1αin skeletal muscle.Conclusion Berberine can improve the expression of PGC-1α,promote the expression of GLUT4 and improve insulin resistance.
