海蛾乙酸乙酯提取物的分离纯化及抗肿瘤活性分析 被引量:2

摘要 通过高效液相色谱技术建立海蛾乙酸乙酯提取物的分离纯化方法,采用新型半制备色谱条件收集组分,获得A1,A2,A3,A4,A5,A6等6个单组分,采用MTT法测定各组分对肿瘤细胞的增殖抑制作用.实验结果表明,A5、A6组分对He La细胞的增殖抑制作用最为显著.流式细胞术Annexin V/PI双染检测细胞凋亡情况发现,组分A5、A6对He La细胞的增殖抑制作用可通过诱导He La凋亡途径实现.对样品进行核磁共振以及质谱分析,初步推测A5为芳香族有机化合物,A6为甾体类或萜类有机化合物. Six single components of A1,A2,A3,A4,A5 and A6 were obtained from ethyl acetate crude extract of Pegasus laternarius Cuvier by means of high performance liquid chromatography(HPLC)technology.Then MTT assay was introduced to determine the anti-tumor activity.It was found that the components A5 and A6 have the most significant inhibition effect on HeLa cell line.The Annexin V/PI staining observed in the flow cytometer showed the inhibition effect of the components A5 and A6 was achieved by inducing apoptosis in HeLa cells.Samples were further analyzed by mass spectrometry and nuclear magnetic resonance,which infered that A5 might be an aromatic molecule and A6 might contain a steroid or terpene residue.
