SHR大鼠肾脏去交感神经与主动脉miRNA相关性分析 被引量:3

摘要 目的:对自发性高血压大鼠(SHR)行肾脏去交感神经术(RSD),检测其降压效果以及血和主动脉组织中miRNAs,探讨SHR大鼠肾脏去交感神经与主动脉miRNA相关性。方法:SHR对照组,SHR手术组,正常大鼠(WKY)对照组和WKY手术组,20只/组,SHR对照组和WKY对照组均为假手术组,SHR手术组和WKY手术组接受RSD。记录所有大鼠术前和术后4、8、16和24周尾动脉血压,采用RT-PCR检测血清miRNAs及24周主动脉组织miRNAs(miR-150、miR-155、miR-124、miR-135a和miR-143/145)。结果:4组大鼠不同时间点的血压、miRNAs均有差别(P<0.05),与SHR对照组比,SHR手术组的收缩压、舒张压和miRNAs较低(P<0.05),同与WKY对照组比,WKY手术组的收缩压和舒张压较低(P<0.05),WKY对照和WKY手术组的miRNAs无统计学意义(P>0.05)。结论:RSD可降低SHR血压,可能与降低主动脉组织和循环血清miRNAs表达有关。 Objective:To study the effect of renal sympathetic denervation(RSD)for spontaneously hypertensive rats(SHR)and the relationship between RSD and expressions of miR-150、miR-155、miR-124、miR-135a and miR-143/145 in aorta artery tissue and peripheral blood.Method:A total of 40 healthy SHR Wistar rats and 40 normal rats(WKY)were divided randomly into operation and sham sub-groups,20 rats in each group:SHR control group,SHR operation group,WKY control group and WKY operation group,SHR control group and WKY control group accepted sham operation,SHR operation group and WKY operation group underwent operation.The miR-150,miR-155,miR-124,miR-135a and miR-143/145 in peripheral blood before and 4,8,16 and 24 weeks after operation,the values of systolic(SBP)and diastolic(DBP)blood pressure of rat tail artery;the above miRNAs in aorta artery tissue by RT-PCR after 24 weeks by killed rats.Results:The blood pressure and miRNAs of 4 groups were different at different time(P<0.05).Compared with the SHR control group,the systolic and diastolic blood pressure and miRNAs of SHR operation group were lower(P<0.05).Compared with the WKY control group,the systolic and diastolic blood pressure and miRNAs of WKY operation group were lower(P<0.05).The miRNAs of WKY control group and WKY operation group had no differences(P>0.05).Conclusion:It is effective and stable for RSD in SHR rats by lowerring miR-150,miR-155,miR-124,miR-135a and miR-143/145 in peripheral blood and aorta artery tissue.
