花青素合成转录因子Vlmyb2遗传转化烟草的研究 被引量:1

摘要 本研究以烟草为材料,利用农杆菌介导法导入花青素合成转录因子Vlmyb2,对抗性植株进行鉴定得到转基因植株。实验结果表明:转Vlmyb2基因烟草植株的花青素含量高于4 mg/g的转基因植株,叶绿素含量反而低,花青素与叶绿素呈现出较为明显的线性反比关系。当花青素含量较低,叶绿素含量反而升高。测定烟草光合速率实验结果表明:转Vlmyb2基因调节花青素结构基因在烟草中超表达会降低植株的光合效率,但降幅并不显著,对植株正常生长没影响。扫描电镜实验结果表明:Vlmyb2转录因子表达量高的株系腺毛密度比对照高了约23%,相反,Vlmyb2转录因子表达量低的株系的比对照低了约15%,Vlmyb2转录因子的表达影响了烟草腺毛的发育。 In this study,tobacco was used as the material to introduce the anthocyanin biosynthesis transcription factor V1myb2 by Agrobacterium-mediated transformation,and the transgenic plants were identified through the identification of resistant plants.The results showed that the anthocyanin content in V1myb2 transgenic plants was higher than that of 4 mg/g transgenic plants,the content of chlorophyll was rather low.Anthocyanin and chlorophyll showed obvious linear inverse relationship;when anthocyanin content is low,chlorophyll content increases instead.Results from the determination of tobacco photosynthesis rate showed that:over-expression of transgenic Vlmyb2 gene regulation of anthocyanin structural genes in tobacco would reduce photosynthetic efficiency,but the decline was not significant and did not affect the normal growth of the plant.The results of scanning electron microscopy showed that the density of glandular hairs in he transgenic plants with high V1myb2 transcription factor was about 23%higher than that of the control.In contrast,the low expression of the V1myb2 transcription factor was about 15%lower than that of the control.The V1myb2 transcription factor expression affects the development of tobacco glandular hairs.
