5种高产洛伐他汀曲霉的筛选和鉴定 被引量:2

Screening and Identifying of 5 Higher-Yield Lovastatin Aspergillus Species

摘要 从不同生境(食品、土壤、空气、有机质等)收集的自然发酵样品中分离纯化获得150株曲霉纯菌株;经薄层层析法(TLC)初筛获得25株产洛伐他汀(Lovastatin)的曲霉菌株;进一步用高效液相色谱法(HPLC)复筛获得5株Lovastatin产量较高的曲霉菌株,产量范围为32~56 mg/L。对筛选的5株曲霉菌株进行形态特征描述和鉴定,依据曲霉菌株菌落特征、显微特征,结合18S r DNA或ITS r DNA基因序列,鉴定这5株菌株分别为棒曲霉(Aspergillus clavatus)、塔宾曲霉(Aspergillus tubingensis)、菌核曲霉(Aspergillus sclerotiorum)、黑曲霉(Aspergillus niger)和米曲霉(Aspergillus oryzae)。 One hundred and fifty Aspergillus strains were obtained by collected,separated and purified from natural fermentation samples food,soil,air and so on.Then 25 Aspergillus strains were screened by assaying Lovastatin with thin layer chromatography(TLC).And 5 Aspergillus strains which produces higher-yield Lovastatin was screened by high performance liquid chromatography(HPLC),with Lovastatin yield range 32~56 mg/L.Based on 18S rDNA or ITS rDNA sequence and morphology characteristics,the 5 Aspergillus strains were identified as Aspergillus clavatus,Aspergillus tubingensis,Aspergillus sclerotiorum,Aspergillus niger,Aspergillus oryzae,respectively.
