血管内皮生长因子及细胞间黏附分子-1在糖尿病大鼠内耳的表达及其临床意义

The expression and clinical significances of vascular endothelial growth factor and intercellular adhesion molecules-1 in inner ear of diabetes mellitus rats

摘要 目的探讨糖尿病大鼠内耳中血管内皮生长因子(vascular endothelial growth factor,VEGF)和细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)的表达水平及其临床意义。方法将48只Wistar大鼠随机分为对照组和糖尿病组,每组各24只。对照组大鼠普通饲养;糖尿病组大鼠高脂、高糖饲养,并腹腔注射链脲佐菌素65 mg/kg,当空腹血糖>16.65 mmol/L时显示造模成功,分别于0、4、8、12周时取出实验大鼠耳蜗进行免疫组化观察。结果 0、4、8、12周,糖尿病组大鼠血管壁厚度均显著大于对照组(P<0.05),两组大鼠VEGF和ICAM-1表达水平差异均具有显著性(P<0.05);VEGF和ICAM-1表达水平呈正相关(r=0.931,P<0.01)。结论糖尿病大鼠内耳血管壁增厚;VEGF和ICAM-1在糖尿病大鼠内耳的表达增强,两者表达水平呈正相关。 Objective The present study was to study the expression of vascular endothelial growth factor(VEGF)and intercellular adhesion molecules-1(ICAM-1)in the inner ear of diabetes rats and their clinical significances.Method 48 Wistar rats were randomly divided into control group and diabetes group,each group had 24 cases,control group rats were feed with ordinary feed;diabetes group rats were feed with fat and sugar food.Moreover,65 mg/kg streptozotocin(intraperitoneal injection)was given to the rats in diabetes group so as to receive the gold of fasting blood glucose>16.65 mmol/L,which showed the success of building mode.On weeks 0,4,8,12,the cochlear lookahead was taken out for immunohistochemical experiment.Result Diabetics group vascular wall thickness were larger than that of control group on weeks 0,4,8,12(P<0.05);the expression levels of VEGF and ICAM-1 in the two groups were significantly different(P<0.05);the expression level of VEGF and ICAM-1 was positively correlated(r=0.931,P<0.01).Conclusion The vascular wall thickness in inner ear of diabetic rats became thicken.The expression of VEGF and ICAM-1 enhanced in diabetic rats inner ear,the VEGF and ICAM-1 in inner ear of diabetic rats,and they are positively corrected.
