摘要 目的探讨外周血单核细胞CD14和toll样受体(TLR4)mRNA的表达与帕金森综合征(PD)的关系。方法前瞻性选取2016年1月至2017年12月在北京潞河医院治疗的PD患者80例,同时选取健康志愿者80例作为对照组。采用RT-PCR技术检测CD14 mRNA和TLR mRNA表达,同时分析CD14 mRNA和TLR4 mRNA表达与PD患者临床特征的关系,CD14 mRNA和TLR mRNA表达的相关性。结果 PD组CD14 mRNA和TLR mRNA相对表达量分别为(1.522±0.344)和(1.417±0.401),明显高于对照组(P<0.05);H-Y分期>2.5期患者CD14 mRNA和TLR4 mRNA相对表达量分别为(1.843±0.312)和(1.722±0.422),明显高于H-Y分期≤2.5期患者(P<0.05);CD14 mRNA和TLR4 mRNA相对表达量与PD患者性别、年龄、病程、运动并发症、服用受体激动剂和左旋多巴类药物无关(P>0.05);CD14 mRNA和TLR mRNA相对表达量呈正相关(P<0.05)。结论 CD14 mRNA和TLR4 mRNA表达与PD患者疾病严重程度有一定关系,提示机体固有免疫可能参与了PD的发生发展。 Objective To investigate the relationship between the expression of CD14 and toll like receptor(TLR4)mRNA in peripheral blood mononuclear cells and Parkinson syndrome(PD).Methods 80 cases of PD patients who were treated from January 2016 to December 2017 were selected,and 80 healthy volunteers were selected as the control group,CD14 mRNA and TLR mRNA expression were detected by RT-PCR technique,and the relationship between the expression of CD14 mRNA and TLR4 mRNA and the clinical characteristics of PD patients were analyzed.The correlation between CD14 mRNA and TLR mRNA expression was analyzed.Results The relative expression of CD14 mRNA and TLR mRNA in PD group was(1.522±0.344)and(1.417±0.401),which were significantly higher than those in the control group(P<0.05).H-Y stage>2.5 patients CD14 mRNA and TLR4 mRNA expression levels were(1.843±0.312)and(1.722±0.422),which were significantly higher than those of H-Y stage≤2.5 patients(P<0.05).The relative expression level of CD14 mRNA and TLR4 mRNA were not related to the gender,age,course of disease,motor complications,taking receptor agonists and L-DOPA drugs in PD patients(P>0.05).The relative expression of CD14 mRNA was positively correlated with TLR mRNA(P<0.05).Conclusion The expression of CD14 mRNA and TLR4 mRNA are related to the severity of the disease in patients with PD,suggesting that the inherent immunity of the body may be involved in the development of PD.
