摘要
目的:探讨抑制EphA2受体对内毒素性急性肺损伤的保护作用。方法:将成年SD大鼠随机分为对照组(C组)、LPS组(L组)和抑制剂组(M组),检测给予LPS后1 h、3 h和6 h血清中炎症细胞因子肿瘤坏死因子(TNF-α)和白介素(IL-6)水平的变化,检测大鼠肺组织湿/干重(W/D)比值,检测肺组织髓过氧化物酶(MPO)活性的变化。结果:与对照组相比,L组在3个时间点TNF-α和IL-6的含量升高(P<0.05);而给予EphA2/Fc后,M组TNF-α和IL-6的含量低于L组(P<0.05);与对照组相比,LPS组肺组织W/D的比值和MPO活性升高(P<0.05),而给予EphA2/Fc后,M组W/D的比值和MPO活性低于L组(P<0.05)。结论:使用抑制剂EphA2/Fc抑制EPh A2受体对内毒素性急性肺损伤有保护作用,EphA2受体可以作为急性肺损伤研究治疗的新靶点。
Objective:To investigate the protective effects of EphA2 inhibition receptor on LPS-induced acute lung injury(ALI).Methods:Adult SD rats were randomly divided into control group(group C),LPS group(group L)and EphA2/Fc group(group M).TNF-αand IL-6 in serum were detected on 1h,3h and 6h after LPS administration.The wet/dry(W/D)ratio was detected and the MPO activity was detected.Results:Compared with control group,the content of TNF-αand IL-6 in group L increased significantly after LPS administration(P<0.05).However,the content of TNF-αand IL-6 in group M was significantly lower than that in group L after EphA2/Fc administration(P<0.05).Compared with control group,the W/D ratio and MPO activity in group L increased significantly(P<0.05).However,the W/D ratio and MPO activity in group M was significantly lower than that in group L after EphA2/Fc administration(P<0.05).Conclusion:EphA2 inhibition receptor has protective effects on LPS-induced ALI.EphA2 receptor would be a new target for the treatment of ALI.
