海地瓜胶原蛋白抗氧化活性肽的酶解工艺优化

Optimization of Enzymatic Hydrolysis of Acaudina Molpadioides Semper Collagen Polypeptides and Analysis of Antioxidant Activity

摘要 以海地瓜为研究对象,以复合蛋白酶为催化剂,以胶原蛋白酶解液DPPH清除率等抗氧化活性为衡量指标,选取底物浓度、酶用量、pH、酶解温度、酶解时间等条件为优化对象,在单因素基础上,进行响应面分析法(RSM),确定最佳抗氧化活性的海地瓜胶原蛋白肽。试验结果表明,海地瓜胶原蛋白酶解最佳条件为底物浓度150g.L-1,酶用量3AIM71,pH7.0,酶解温度53.5°C,酶解时间5.0h,此时DPPH清除率为(76.63±0.86)%,与模型预测值相符,本方法制备的胶原蛋白肽具有较强的抗氧化能力。 Protamex was used as catalyzer to hydrolyze the Acaudina molpadioides Semper,and the removal efficiency of DPPH was selected as response parameter.Based on the single factor analysis,the substrate concentration,enzyme addition,pH,enzymolysis temperature,and enzymolysis time were screened and optimized using response surface methodology(RSM)to determine the optimal DPPH Hydroxyl radical scavenging rate.The results showed that the optimal conditions are as follows:150 g.L-1 of substrate concentration,3 AU.g_1 of enzyme addition,pH of 7.0,temperature at 53.5℃,and enzymolysis for 5 h.The DPPH scavenging rate is 76.63±0.86%under the optimal conditions,which correspond with the prediction model.Our data indicated that the antioxidant activity of Acaudina molpadioides Semper collagen have strong antioxidant capacity.
