摘要
目的研究辛伐他汀预处理保护大脑脑缺血再灌注损伤。方法选择2016年10月—2017年5月在该院观察的30只健康的雄性大鼠作为该文的研究对象,将大鼠随机分为3组,每组10只大鼠,分别为辛伐他汀预处理组(A组)、模型加溶媒组(B组)和假手术组(C组),使用线栓法制作大鼠大脑动脉缺血再灌注模型,并且进行2 h的再灌注,24h之后对大鼠的神经功能进行评分,并且将大鼠断头取脑,对大鼠的凋亡细胞数、脑梗死体积和Fas表达进行测定。结果 B组大鼠的脑梗死体积为(36.2±54.20)%,A组大鼠的脑梗死体积为(23.2±37.50)%,和B组相比,A组的脑梗死体积有明显的缩小;B组的神经功能缺损为(2.80±0.42)分,A组的神经功能缺损为(1.62±0.23)分,A组神经功能缺损评分比B组的改善明显,数据差异有统计学意义(P<0.05);A组、B组的Fas表达分别为(31.25±2.65)%、(65.25±4.25)%,A组和B组的凋亡细胞数量为(30.21±3.14)%、(44.26±4.12)%,以此表示A组脑组织Fas表达和凋亡的细胞数量比B组降低明显,数据差异有统计学意义(P<0.05)。结论辛伐他汀能够保护大鼠脑缺血再灌注的损伤,通过对脑组织Fas表达的抑制降低细胞的凋亡。
Objective To study the simvastatin pretreatment in protecting the cerebral ischemia reperfusion injury in rats.Methods 30 cases of healthy male rats observed in our department from October 2016 to May 2017 were selected as the research objects and randomly divided into three groups with 10 cases in each,including the simvastatin pretreatment group(A group),model and solution medium group(B group)and sham-operation group(C group),and the cerebral artery ischemia reperfusion model of rats was made by the suture-occluded method for two-hour reperfusion,and the nerve function was marked after 24 h,and the number of apoptotic cells,cerebral infarct volume and Fas expression of rats were measured.Results The cerebral infarct volume in the group A was obviously shortened compared with that in the group B,[(23.2±37.50)%vs(36.2±54.20)%],and the nerve function defect in the group A was obviously improved compared with that in the group B,[(1.62±0.23)points vs(2.80±0.42)points],and the difference was statically significant(P<0.05),and the Fas expression in the group A and in the group B was respectively(31.25±2.65)%and(65.25±4.25)%,and the number of apoptotic cell in the group A and in the group B was respectively(30.21±3.14)%and(44.26±4.12)%,therefore,the Fas expression and number of apoptotic cells in the group A obviously decreased compared with those in the group B,and the differences were statistically significant(P<0.05).Conclusion The simvastatin can protect the cerebral ischemia reperfusion injury in rats,and it can reduce the cell apoptosis by restraining the Fas expression of cerebral tissues.
