摘要
目的研究性别及年龄对原发性高血压(EH)大鼠炎症因子的影响。方法采用6月龄及12月龄原发性高血压大鼠(SHR),用无创尾动脉测压系统测定血压,用放射免疫分析法测定血清肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)浓度,应用实时荧光定量PCR(real-time PCR,RT-PCR)技术检测肾脏组织中的TNF-α,IL-6的mRNA。结果雄性SHR(SHR-M)大鼠血清IL-6高于同龄的雌性(SHR-F)大鼠(P<0.05),12月龄SHR(SHR-12m)大鼠高于6月龄SHR(SHR-6m)大鼠(P<0.05)。SHR-6m大鼠的血清TNF-α不存在性别差异,SHR-12m-M大鼠高于SHR-12m-F大鼠(P<0.05);SHR-12m大鼠高于SHR-6m大鼠(P<0.05)。SHR-6m-F大鼠肾组织中IL-6mRNA的表达低于SHR-6m-M大鼠(P<0.05),SHR-12m大鼠高于同性别SHR-6m大鼠(P<0.05)。SHR-6m-F大鼠和SHR-12m-F组肾脏组织中的TNF-α的mRNA的表达低于同龄雄性大鼠(P<0.05),无年龄因素的影响。结论高血压性炎症反应存在性别差异性,并随着高血压发病时间的延长炎症反应的加重。
Objective To study the effects of gender and age on inflammatory factors in spontaneous hypertensive rats(SHR).Methods Blood pressure was measured by tail-cuff method(Non-invasive tail artery blood pressure measuring device),the concentration of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in serum were detected by radioimmunoassay in 6 months and 12 months spontaneous hypertensive rats(SHR).The mRNA levels of TNF-αand IL-6 in kidney tissue were determined using quantitative real-time polymerase chain reaction.Results Compared with SHR-Male group,serum IL-6 was significantly higher in the same age SHR-Female group(P<0.05);compared with SHR-6m group,serum IL-6 was significantly higher in the same gender SHR-12m group(P<0.05).Serum TNF-αwas no significantly change in the SHR-6m group,compared with the SHR-12m-Male group,serum TNF-αwas significantly lower in the SHR-12m-Female group(P<0.05).mRNA expression of IL-6 in the kidney in the SHR-6m-Male group and SHR-12m-Male group were significantly higher than that in the same age females group(P<0.05).Compared with the female group,mRNA levels of TNF-αin the kidney of the SHR-6m group and SHR-12m group were significantly increased(P<0.05),there was no age factor change in the same gender group.Conclusion There is a gender difference in the inflammation during the development of hypertension,and the severity of inflammation increases with the time prcessing of onset of hypertension.
