摘要 目的探讨黄芩苷对前列腺癌细胞生物学行为的影响。方法通过CCK-8细胞活性实验检测不同浓度黄芩苷对前列腺癌PC3细胞活性的影响;MTT细胞增殖实验检测黄芩苷对前列腺癌PC3细胞增殖能力的影响;流式细胞术检测黄芩苷对前列腺癌细胞凋亡行为的影响和细胞内活性氧(ROS)表达水平的变化。结果黄芩苷浓度为25 mg/L时对前列腺癌PC3细胞抑制率最高(P<0.05)。在72和96 h时,黄芩苷组的前列腺癌PC3细胞增殖率低于对照组(P<0.05)。黄芩苷组的前列腺癌PC3细胞凋亡率和细胞中ROS表达水平高于对照组(P<0.05)。结论合适浓度(25 mg/L)的黄芩苷可以抑制前列腺癌细胞的增殖,并促进其凋亡,抑制前列腺癌细胞的生物学行为。 Objective To investigate effect of Baicalin on biological behaviour of prostate cancer cells.Methods CCK-8 cell activity test was used to detect the effects of different concentrations of Baicalin on PC3 cytoactive in prostate cancer.Methyl thiazolyl tetrazolium(MTT)cell proliferation test was used to detect the effect of Baicalin on PC3 cell proliferation in prostate cancer.Effect of Baicalin on apoptosis of prostate cancer cells and changes of intracellular reactive oxygen species(ROS)expression were detected by flow cytometry.Results When Baicalin concentration was 25 mg/L,the inhibition rate on PC3 cells was the highest(P<0.05).At 72 and 96 h after staining,proliferation rate of PC3 cells in Baicalin group was significantly lower than that in control group(P<0.05).Apoptosis rate and ROS expression in PC3 cells in Baicalin group were significantly higher than that in control group(P<0.05).Conclusion Suitable concentration(25 mg/L)of Baicalin may inhibit proliferation of prostate cancer cells,promote its apoptosis and inhibit biological behaviour of prostate cancer cells.
