头穴丛刺法对脑缺血再灌注大鼠脑组织BDNF、p75NTR表达的影响 被引量:13

摘要 目的:研究头穴丛刺法对脑缺血再灌注(MCAO/R)模型大鼠脑组织中脑源性神经营养因子(BDNF)和神经营养素受体p75(p75NTR)表达水平的影响,探讨其治疗脑缺血再灌注损伤的机制。方法:采用SD大鼠线栓法复制MCAO/R模型,随机分为模型对照组和针刺组,另设空白对照组和假手术组,每组各12只。针刺组大鼠采用头穴丛刺法进行针刺干预,在再灌注后2 h即开始针刺,每12 h针1次,连续针刺10次。各组大鼠在末次针刺干预后30 min,取出脑组织,分别采用免疫组化法、WB法和Realtime-PCR法检测缺血区脑组织中BDNF及p75NTR蛋白与基因的表达。结果:与空白对照组相比,模型对照组大鼠缺血区脑组织内BDNF及p75NTR基因和蛋白表达均显著增加(P<0.01);与模型对照组相比,针刺组大鼠缺血区脑组织中BDNF的蛋白与基因表达水平均明显升高(P<0.01),而p75NTR蛋白和基因的表达均明显著降低(P<0.01)。结论:头穴丛刺法可能通过上调缺血区脑组织中BDNF的蛋白与基因表达水平、下调p75NTR的蛋白与基因表达水平从而起到对脑组织的神经保护作用。 Objective:To study the effect of scalp cluster needling on the expressions of brain-derived neurotrophic factor(BDNF)and p75 neurotrophin receptors(p75NTR)in brain tissue in MCAO/R(middle cerebral artery occlusion/reperfusion)rats,thus to explore the mechanism of the therapy treating cerebral ischemia reperfusion(CIR).Methods:Focal cerebral ischemia was induced by middle cerebral artery occlusion method using a monofilament thread in SD rats.The model rats were randomly divided into the model group and the acupuncture group;also,rats in the blank group and sham-operated group were taken as the control,12 rats in each group.The acupuncture group was treated with sclap cluster needling,which started after 2 h of the reperfusion,once per 12 h,10 times in a row.The brain tissues of the rats in each group were removed 30 minutes after the last acupuncture intervention.The protein and gene expressions of BDNF and p75NTR were measured by the methods of immunohistochemisty,Western blot and PCR.Results:The expressions of BDNF and p75NTR were significantly increased in the model group than those in the blank group(P<0.01).The expression of BDNF was obviously increased,and the expression of p75NTR were markedly decreased in the acupuncture group compared to those in the model group(P<0.01).Conclusion:The therapy of scalp cluster needling can up-regulate BDNF expression and down-regulate p75NTR expression to play a neuroprotective role in brain tissue.
