摘要
目的:通过观察TLR-4蛋白表达的变化,研究头穴透刺治疗脑缺血再灌注损伤所产生的抗炎作用机制。方法:随机将70只SD大鼠分为假手术组、模型组和头针组。采用改良线栓法建立右侧大脑中动脉阻断的缺血再灌注(MCAO/R)大鼠模型。头针组采用头穴透刺法:右侧百会透曲鬓穴。采用免疫组化法检测各组大鼠缺血侧脑组织中TLR-4蛋白的定性表达情况;采用Western blot法检测各组大鼠缺血侧脑组织中TLR-4蛋白的定量表达情况。结果:模型组和头针组不同时间点缺血脑组织中TLR-4蛋白含量均较假手术组明显升高(P<0.05);但头针组缺血脑组织中TLR-4蛋白含量均较同时间点模型组明显降低(P<0.05)。结论:早期头针治疗可抑制脑缺血再灌注后缺血侧脑组织中TLR-4的表达。TLR-4表达水平下调可能是头穴透刺法拮抗脑缺血再灌注损伤的分子机制之一。
Objective:To study the anti-inflammatory mechanism of scalp point-to-point needling in the treatment of cerebral ischemia reperfusion injury(CIRI),by observing the expression of TLR-4 protein.Methods:70 SD rats were randomly divided into the sham-operated group,the model group,and the scalp acupuncture group.Middle cerebral artery occlusion/reperfusion(MCAO/R)model was established in rats with modified suture-occluded method.Left needling of Baihui(GV20)-to-Qubin(GB7)was applied in the rats of the scalp acupuncture group.The qualitative expression of TLR-4 protein was detected by immunohistochemistry method in ischemic brain tissue;the quantitative expression of TLR-4 protein was detected by the western blot method.Results:The content of TLR-4 protein was significantly higher in the model group and in the scalp acupuncture group than that of the sham-operated group at different time point in the ischemic brain tissue(P<0.05);however,the content of TLR-4 protein was significantly lower in the scalp acupuncture group than that of the model group at the same time point(P<0.05).Conclusion:Early scalp needling intervention can inhibit the expression of TLR-4 protein of in MCAO/R rats,which may be the anti-inflammatory mechanism of scalp point-to-point needling in the treatment of CIRI.
