基于半仿生及体外抗肿瘤细胞株实验技术对土鳖虫最佳用酶的筛选 被引量:2

Screening Optimized Hydrolase of Eupolyphaga by Using Semi-Bionic Technology and MTT Method of Anti-tumors in Vitro

摘要 以半仿生技术结合体外抗肿瘤细胞实验,筛选出土鳖虫的最佳用酶。方法:以土鳖虫为底物,用胃蛋白酶、胰蛋白酶、胰凝乳蛋白酶和弹性蛋白酶进行酶解,同时以不加酶的水提液为对照组,测定每组的水解度,采用MTT法,以对肿瘤细胞株的抑制率为指标,分别对土鳖虫的最佳用酶进行筛选。水解度从高到低的顺序依次为弹性蛋白酶、胰蛋白酶、胃蛋白酶和胰凝乳蛋白酶,它们的水解度分别为9.95%、6.76%、4.94%、2.29%;不同酶解液对不同肿瘤细胞抑制效果不同。土鳖虫酶解液对肝癌细胞的最佳用酶是胰蛋白酶、弹性蛋白酶、胰凝乳蛋白酶;对喉癌细胞的最佳用酶是胰蛋白酶和弹性蛋白酶。胃蛋白酶对两个肿瘤细胞的抑制效果最弱可忽略不计。 To screen out the Optimized hydrolase of Eupolyphaga by Using Semi-Bionic Technology and MTT method of anti-tumor in vitro.Using Eupolyphaga as substrate,pepsin,trypsin,chymotrypsin and elastase were used for enzymolysis,and the water extract without enzyme was used as the control group to determine the degree of hydrolysis of each group.The inhibitory ratios of MTT assay for the tumor Cell lines were regarded as the index,and then confirmed the Optimized hydrolase of Eupolyphaga.The degrees of hydrolysis showed that the order from high to low was elastase,trypsin,pepsin and chymotrypsin,and their degrees of hydrolysis were 9.95%,6.76%,4.94%and 2.29%respectively.Different enzymatic hydrolysates had not same inhibitory effects for each tumor cell lines.Conclusion:The optimized hydrolase of Eupolyphaga for liver cancer cell lines were trypsin,elastase and chymotrypsin;trypsin and elastase for laryngeal cancer cell lines;Pepsin had the weakest inhibitory effect on both tumor cells,so it could be negligible.
