摘要
目的建立瓜子金皂苷的指纹图谱,对12批瓜子金皂苷提取物进行质量评价,为瓜子金药材质量的控制提供依据。方法采用超高效液相法,Acquity UPLCHSS T3(100 mm×2.1 mm,1.8μm)色谱柱,以乙腈-水为流动相梯度洗脱,流速为0.4 m L/min,柱温为30℃,检测波长为230 nm,建立瓜子金皂苷UPLC指纹图谱及对12批药材进行相似度评价,并采用SPSS22.0软件进行聚类分析及主成分分析。结果 12批不同产地不同批次的瓜子金药材皂苷提取物标定了21个共有峰,相似度在0.5以上;聚类分析结果显示,12批药材提取物分为3类,与产地有一定关系;主成分分析结果显示,春季采摘且存储时间越短的药材得分越高。结论本方法稳定性好,方法简便,所建立的指纹图谱对进一步研究瓜子金皂苷药效物质基础有重要意义,也为瓜子金的质量控制与评价提供了参考。
Objective To establish a fingerprint chromatogram of total saponins from Polygala japonica Houtt by ultra performance liquid chromatography(UPLC),and to evaluate the quality of 12 batches of Polygala japonica Houtt saponins extract and provide a scientific basis for its quality standard.Methods The column of Acquity UPLC HSS T3(100 mm×2.1 mm,1.8μm)was eluted with a mobile phase of acetonitrile-water at 0.4 mL/min detected with the wavelength of 230 nm.The UPLC fingerprinting of melon seeds was established.The similarity of 12 batches of medicinal materials was evaluated by the similarity evaluation system of chromatographic fingerprints of Chinese traditional medicine.The obtained chromatographic data were analyzed by cluster analysis and principal component analysis using SPSS 22.0 software.Results 21 common peaks were screened from twelve batches of different regions of Polygala japonica Houtt.The similarity was above 0.5.By clustering analysis,the extracts were divided into three categories and the results showed that they had a certain relationship with the origin.The score of the extract was calculated by principal component analysis.The results showed that the score of spring-picking and short storage time was higher.Conclusion The method is stable and simple,and the established fingerprint is of great significance for further study on the substance basis of melon seed saponins,and also for the quality control and evaluation of Polygala japonica Houtt.
