摘要
本文以α-葡萄糖转苷酶为研究对象,研究了啤酒酵母自溶物替代商业酵母膏作为培养基对发酵产酶的影响,通过单因素试验得到最适的酵母自溶物的添加量为120 m L/L。在此基础上,以发酵酶活力为指标,通过正交试验法对黑曲霉发酵产酶的条件进行了优化,结果表明,产酶最佳条件为发酵温度30℃,发酵初始p H 6.5,发酵时间120 h,摇床转速200 r/min,通过验证试验,得出α-葡萄糖转苷酶的酶活力可达200 U/m L。
The effect of yeast autolysate substitute commercial yeast extract onα-Transglucosidase fermentation was studied,the results showed that the addition of the yeast autolysis was 120 mL/L by single factor experiment.On this basis,the fermentation conditions for producingα-Transglucosidase from Aspergillus niger were optimized by orthogonal test method.The optimum conditions for fermentation were 30℃,at pH 6.0 and 200 r/min for 48 h.Fermentation conditions were carried out in a 5 L fermenter to obtain the activity ofα-Transglucosidase as high as 200 U/mL.
