摘要
目的研究miR-29a对胎盘滋养细胞增殖和凋亡的影响及潜在机制。方法采用Real-time PCR技术检测miR-29a和MCL1 mRNA的表达情况,CKK-8法检测miR-29a对人绒毛外滋养细胞增殖的影响;流式细胞术检测miR-29a对滋养细胞凋亡的影响。结果过表达miR-29a后人绒毛外滋养细胞的增殖能力降低、凋亡水平增加。Real-time PCR和Western blot结果显示,过表达miR-29a抑制MCL1的mRNA表达,下调MCL1蛋白水平。干扰miR-29a后,人绒毛外滋养细胞的增殖能力升高,凋亡水平降低,并且MCL1的mRNA和蛋白水平升高。结论 miR-29a通过下调MCL1蛋白水平抑制人绒毛外滋养细胞增殖,促进滋养细胞凋亡。
Objective To study the effects and underlying mechanisms of miR-29a on the proliferation and apoptosis of placental trophoblast cells.Methods The expression of miR-29a and MCL1 mRNA was measured by RT-PCR.The effect of miR-29a on the proliferation of human trophoblast cells was measured by the CKK-8 assay.Flow cytometry was used to detect the effect of miR-29a on apoptosis.The protein levels of were detected by western blot.Results The proliferation of human extravillous trophoblast cells was decreased after overexpression of miR-29a.Apoptosis of human extravillous trophoblast cells was upregulated with the overexpression of miR-29a.The results of real-time PCR and western blots showed that overexpression of miR-29a inhibited MCL1 mRNA and protein levels.After interference with miR-29a,human villous trophoblast cell proliferation increased,the level of apoptosis decreased,and MCL1 mRNA and protein levels increased.Conclusion miR-29a inhibite the proliferation of trophoblasts outside of human villi,down-regulate protein levels of MCL1 and promot apoptosis in trophoblasts.
