响应面法优化金莲花中荭草苷和牡荆苷提取工艺 被引量:3

Extraction of Orientin and Vitexin from Trollius chinensis Bunge Using Response Surface Methodology

摘要 采用中心组合-响应面法(CCD-RSM)对金莲花中荭草苷和牡荆苷提取工艺进行优化,以期为金莲花的开发提供一定的参考。通过单因素试验确定超声提取影响因素与水平,并以此为基础采用中心组合-响应面法研究了提取时间、提取温度和料液比对金莲花中荭草苷和牡荆苷提取率的影响,确定超声提取金莲花中荭草苷和牡荆苷的最佳工艺参数为:提取时间60min,提取温度60℃,料液比1∶12 g·m L-1。验证试验表明,荭草苷和牡荆苷的总提取率为1.89%,与预测的最大响应值十分接近,说明回归模型的拟合度较高,可用于金莲花中荭草苷和牡荆苷的提取,为金莲花的开发利用提供一定的理论参考。 The ultrasonic-assisted extraction conditions for orientin and vitexin from Trollius chinensis Bunge were optimized by the central composite design and response surface methodology,to provide some reference for the development and utilization of Trollius chinensis Bunge.Through the single factor tests,the effects of ultrasonic extraction factors and levels were determined;and these were the basis for the central composite design and response surface methodology to research the functions of extraction time,extraction temperature and solid-liquid ratio on the extraction rate of orientin and vitexin from Trollius chinensis Bunge.The results showed that the optimum conditions were:solid-liquid ratio,1∶12 g·mL-1;extraction time,60 min and extraction temperature,60℃.The yield of orientin and vitexin was up to 1.89%under the optimal extraction conditions.Validation experiments showed that the model equation can well predict the result of the experiment with good fitting,can be used for the extraction of orientin and vitexin from Trollius chinensis Bunge,which provides a theoretical reference for the development and utilization of Trollius chinensis Bunge.
