摘要
目的研究蒺藜的甾体皂苷成分TTS-12对于耐药皮肤癣菌的抗真菌作用。方法用含体外浓度递增法对真菌进行培养,分别诱导红色毛癣菌对氟康唑和酮康唑的耐药性,用微量液基稀释法评价菌株的耐药性、稳定性及对TTS-12的敏感度。结果经过诱导后,耐氟康唑菌株对氟康唑的最小抑菌浓度(MIC)>128μg·m L-1,耐酮康唑菌株对酮康唑的最小抑菌浓度>64μg·m L-1,且传代三次后耐药性相对稳定,TTS-12对氟康唑耐药菌株的最小抑菌浓度范围在4~16μg·m L-1,对酮康唑耐药菌株的最小抑菌浓度范围在4~8μg·m L-1。结论经过浓度递增方式的培养能够成功诱导红色毛癣菌耐氟康唑和酮康唑的菌株,并且TTS-12对两种耐药菌株有良好的生长抑制作用。
Objective To study the antifungal activity of TTS-12,a kind of steroid saponin constituent in caltrop,against drug-resistant fungus.Methods To induce resistance of Trichophyton rubrum against fluconazole and ketoconazole by progressive concentrations cultivation in vitro.Micro-broth dilution assay was adopted to evaluated the drug-resistance level,stability and sensibility of fungus to TTS-12.Results Fluconazole-resistant Trichophyton rubrum(MIC>128μg·mL-1)and ketoconazole-resistant Trichophyton rubrum(MIC>64μg·mL-1)had been induced.Drug-resistance stability can be kept in three generations.The MIC range of TTS-12 against fluconazole-resistant Trichophyton rubrum was 4~16μg·mL-1.The MIC range of TTS-12 against ketoconazole-resistant Trichophyton rubrum was 4~8μg·mL-1.Conclusion Fluconazole-resistant and ketoconazole-resistant Trichophyton rubrum can be induced by progressive concentrations cultivation in vitro.TTS-12 showed good activity of growth inhibition for two drugs-resistant strains.
