摘要
研究并比较木犀草素及其糖苷木犀草苷的抗炎活性.角叉菜胶所致大鼠足跖肿胀,醋酸致小鼠腹腔毛细血管通透性增加等体内实验表明木犀草素及木犀草苷具有一定的抗炎活性.木犀草素5 mg/kg组在1 h时对角叉菜胶所致大鼠足跖肿胀具有一定的抑制作用,而木犀草苷无明显的抑制作用;木犀草素20 mg/kg组对醋酸致小鼠腹腔毛细血管通透性增加有显著抑制作用,且作用强度优于木犀草苷20 mg/kg组.进一步以脂多糖(LPS)诱导的小鼠巨噬细胞RAW 264.7细胞株为炎症反应模型,采用MTT法、Griess法、Western blot法考察木犀草素和木犀草苷的抗炎活性并对比二者之间的活性差异.结果显示,木犀草素对LPS诱导RAW 264.7产生炎症介质NO和炎性蛋白iNOS、COX-2蛋白高表达表现出明显的抑制作用;木犀草苷对LPS诱导RAW 264.7细胞iNOS蛋白高表达具有抑制作用,以上实验结果表明木犀草素和木犀草苷具有显著抗炎活性,且木犀草素的抗炎活性在一定程度上优于木犀草苷.
The objective of this study is to compare the anti-inflammatory effects of luteolin and luteoloside.The in vivo experimental results show that luteolin and luteoloside have some anti-inflammatory activity.5 mg/kg of luteolin exhibits inhibitory activities on carrageenan-induced paw edema in rats at 1 h.However,luteoloside doesn′t exhibit inhibitory activities at the same dosage.20 mg/kg of luteoloside exerts significant inhibitory activity on acetic acid-induced capillary permeability in mice,with better effect than that of luteoloside at the same dosage.The extrinsic inflammatory model is established by LPS activated RAW 264.7 cells.In order to evaluate the anti-inflammatory effects and to compare the different activities of these two compounds,cell viability assay,Griess assay and Western blot methods are used.The results show that luteolin exerts significant inhibitory activity on the inflammatory media NO,the expression of inflammatory proteins iNOS and COX-2 induced by LPS,whereas luteoloside exhibits significant inhibitory activities on the iNOS protein expression induced by LPS.The above results imply that luteolin and luteoloside have significant anti-inflammatory activities and that the activities of luteolin are superior to luteoloside in some degree.
