摘要
目的探讨食管鳞癌细胞X-Ray照射后miR-126、miR-21、miR-101的表达情况及其与DNA损伤修复基因DNA-PKcs的关系。方法实时定量PCR检测不同剂量X-Ray照射EC109食管鳞癌细胞株24 h后,miR-126、miR-21、miR-101、DNA-PKcs mRNA和蛋白的表达。结果 X-Ray照射EC109后,miR-126、miR-101和DNA-PKcs mRNA表达升高(P<0.05)。放射照射后DNA-PKcs蛋白水平有升高趋势,但差异无统计学意义(P>0.05)。其中miR-21与DNA-PKcs mRNA表达呈负相关。3个miRNAs表达和DNAPKcs蛋白表达无明显相关性。结论 X-Ray照射后,食管鳞癌细胞系EC109的miR-126和miR-101表达上调,DNA-PKcs mRNA表达上调,miR-21与DNA-PKcs mRNA表达呈负相关。
To investigate the expressions of miR-126,miR-21 and miR-101 after X-Ray radiation in esophageal squamous cell carcinoma and the relationship with the expression of DNA-PKcs.Methods After 24 hours treated with different doses radiation of X-Ray in EC109 cells,real time quantitive PCR was used to detect the expressions of miR-126,miR-21,miR-101 and DNA-PKcs mRNA.The expression of DNA-PKcs protein was detected by Western blotting.Results After X-Ray exposure to EC109,the expressions of miR-126,miR-101 and DNA-PKcs mRNA were increased(P<0.05).The expression of DNA-PKcs protein also was increased,but there was no significant difference(P>0.05).The expression of miR-21 was negatively correlated with the expression of DNA-PKcs mRNA.The expressions of miR-126,miR-101,miR-21 proteins were not related with the expression of DNA-PKcs protein.Conclusion After X-Ray irradiation,the expressions of DNA-PKcs mRNA,miR-126 and miR-101 in esophageal squamous cell carcinoma cell line EC109 are up-regulated.Expression of miR-21 is negatively correlated with the expression of DNA-PKcs mRNA.
