山药多糖的提取及其对糖尿病小鼠的影响研究 被引量:24

摘要 目的:研究从山药中提取多糖的方法,以及山药多糖对糖尿病小鼠糖脂代谢紊乱的改善作用。方法:山药粉末经超声辅助提取法得到山药粗多糖,用苯酚-浓硫酸比色法测多糖含量。采用腹腔注射四氧嘧啶的方法构建小鼠糖尿病模型,以格列本脲为对照,观察连续灌胃14天不同剂量山药多糖对糖尿病小鼠血糖(GLU)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白-胆固醇(LDL-C)的含量及小鼠体重的影响。结果:490nm为样品的吸收波长,线形回归方程为:Y=0.0054X-0.0044,相关系数R=0.9990(n=5);山药粗多糖的提取率为3.90%,总糖含量为36.72%;山药多糖可有效改善糖尿病小鼠的血糖和血脂代谢紊乱情况,可显著降低糖尿病小鼠体内的血糖(GLU)、总胆固醇(TC)、甘油三酯(TG)及低密度脂蛋白-胆固醇(LDL-C),缓解体重减轻的状况,效果与格列本脲相当。结论:山药多糖具有有效调节糖尿病小鼠糖脂代谢紊乱的作用。 Objective:To study the extraction of yam polysaccharide and its effects on glucose and lipid metabolism in diabetic mice.Method:The content of polysaccharide was determined by ultrasonic-assisted extraction of yam powder.The content was de-tected with Phenol-concentrated sulfuric acid colorimetry.Alloxan was administered intraperitoneally to establish diabetic models in mice.After the success of modeling,the changes of GLU,TC,TG,LDL-G and body mass of mice were examined.Results:The sam-ples'UV absorption was 490run,linear regression equation:Y=0.0054X-0.0044,correlation coefficient R=0.9990(n=5);the extraction rate of yam polysaccharide was 3.90%,total sugar content was 36.72%.Blood glucose and blood lipid levels of diabetic mice were sig-nificantly ameliorated by administration of yam polysaccharide.Compared with diabetic control mice,yam polysaccharide significandy decreased GLU,TC,TG and LDL-G levels of mice,and alleviated the situation of weight loss;the effect is equivalent to glibenclamide.Conclusion:Yam polysaccharide has the effect of regulating the disorder of glucose and lipid metabolism in diabetic mice.
