摘要
目的建立百合指标性成分含量测定及其特征图谱质量表征关联分析方法,以有效、精准地评价百合药材质量。方法采用HPLC-PDA法对16批百合药材中的酚酸类指标性成分(阿魏酸)和皂苷类指标性成分(薯蓣皂苷)进行含量测定,建立百合特征图谱,同时基于特征图谱中特征峰的梳理及其所属化学类型对16批百合药材进行质的表征;基于特征图谱中指标性成分阿魏酸、薯蓣皂苷及酚酸类(以阿魏酸为表征)、皂苷类(以薯蓣皂苷为表征)的含量和峰面积分别对16批百合药材进行量的表征,并基于基准药材将16批百合药材质与量的表征结果进行关联性分析。结果以批号16为基准药材,整体得出:批号10、16、8、4、7、9指标性成分总体含量较高,批号9、4、8、3、14、10关联性最高,综合评价得出批号10、16、8、4、7、9、3、14优良度居前。结论所建立的百合指标性成分含量测定方法简单、方便,构建的质量表征关联分析模式可用于分析百合药材质量与应用有效性,为今后精准地评价百合质量夯实基础。
Objective To establish the content of the index components and quality representation and correlation analysis of Lilium brownie,to evaluation of the quality of it effectively and accurately.Methods HPLC-PDA method was used to determine the contents of ferulic acid and dioscin in Lilium brownii slices,and specific chromatogram of Lilium brownii was established.The quality of 16 batches of Lilium brownii pieces was represented according to the number and chemical type of characteristic peaks on characteristic spectrum.The quantitative representation of the 16 batches of Lilium brownii was based on the contents and peaks area of characteristic index components,wihch including Phenolic acids(represented by ferulic acid),Saponin(represented by dioscin).Correlation analysis of characterization result of quality and quantity was performed.Results In 16 batches of Lilium brownii,batch number 16 was regarded as the reference material,the indicative components in batch number 10,16,8,4,7 and 9 was higher,while the batch number 9,4,8,3,14,10 had high relevance with the reference piece(No.16).The comprehensive evaluation showed that the good degrees of batch number 10,16,8,4,7,9,3 and 14 wer in the front.Conclusion The established method is simple and accurate.The quality analysis and correlation analysis model can be used to analyze the quality and application of Lilium brownii,and lay a solid foundation for the accurate evaluation of Lilium brownii quality in the future.
