摘要
本文以中国西北常绿树种的木醋液为研究对象,采用酸碱萃取法对木醋液中的酚类物质进行萃取,将得到的样品于硅胶柱色谱进行分离,结合TLC得到3个组分,测定其总酚含量,组分2的总酚含量最高,为1 265.261 mg/g,3个组分的总酚含量均高于木醋液原液。3个组分的抗氧化活性实验结果表明:组分2(IC_(50)=6.06μg/m L)对DPPH自由基清除活性最高;在ABTS实验中,组分3(IC_(50)=15.88μg/m L)对ABTS自由基具有较好的清除活性,其次为组分2(IC_(50)=16.03μg/m L)。因此,硅胶柱色谱分离得到的组分2具有进一步研究的潜力。该实验为木醋液中抗氧化活性成分进一步的研究提供理论依据。
Taking wood vinegar as the study object,phenolic compounds in wood vinegar was extracted by acid–base method,the crude extract was separated by silica gel column chromatography and 3 fractions were obtained combined with TLC.Total phenolic content of the fractions were determined,the total phenolic content of the fraction 2 reached 1 265.261 mg/g,and the total phenolic content of the 3 fractions was higher than that of wood vinegar stock solution.In DPPH assay,the fraction 2(IC50=6.06μg/mL)showed the most potent antioxidant activity;in ABTS assay,the fraction 3(IC50=15.88μg/mL)showed more antioxidant activity,followed by fraction 2(IC50=16.03μg/mL).Therefore,the fraction 2 and 3 are potential for further study,and the assays will provide theoretical basis for further study on the antioxidant bioactive compounds from wood vinegar.
