大豆结瘤相关性状的Meta分析及候选基因挖掘 被引量:3

摘要 氮元素是植物生长所需的重要营养元素之一,生物固氮是大豆生长所需氮元素的重要来源。结瘤数目与重量等是研究共生结瘤的主要性状,对共生结瘤相关性状进行基因定位具有重要的意义。鉴于根瘤相关性状的QTL定位结果比较分散,需要选择合适的公共图谱并整合前人结果,将其真正应用于实践中。故对大豆结瘤相关性状的47个QTL进行Meta分析,得到2个"通用QTL",并将得到的"通用QTL"进行大豆基因组注释,在置信区间内找到8个可能与调节大豆结瘤相关的基因。通过转录表达分析发现,有6个大豆基因受根瘤菌诱导表达,其中有4个基因表达变化幅度较大,并且在绥农14与野生豆ZYD00006中表达均有差异;另2个基因未检测到信号。进一步的蛋白结构域分析发现,这些基因分别含有糖基水解酶、酪氨酸蛋白激酶、亮氨酸重复序列型类受体蛋白激酶和钙钠交换蛋白等结构域,结合已有研究结果表明这些差异表达基因编码蛋白的结构域均与植物抗病、植物免疫反应有关,这为研究大豆与根瘤菌间的共生关系提供理论基础。 Nitrogen is one of the important nutrient elements for the growth of plant.Biological nitrogen fixation is an important alternative to the nitrogen fertilizer in soybean production.The nodule number and weight are the main properties of symbiotic nodulation.The gene mapping of the relevant traits in symbiotic nodulation has great value for research.As the results of QTL location of root nodules were scattered,to integrate the known location result to an appropriate public reference map is required,then the results could be used in practice.Thus,this study aimed at the Meta-analysis of 47 QTLs of relevant traits in soybean nodulation.We got two general QTLs,and found eight genes in the located region underlying soybean nodulation trait.After transcriptional expression analysis,we found that 6 genes were induced by rhizobium,4 of them had a large range transcripted variation,and 3 were significant difference between suinong 14 and wild bean ZYD00006.The signals of two other genes were not detected in root.These genes contain structural domains such as glycemic hydrolase,tyrosine kinase,leucine repetitive sequence type receptor protein kinase,and calcium sodium exchange protein through the further analysis of the protein structure domain.Together with the existed previous results,we found that transcriptional difference expressed genes encode conserved protein domains associated with plant resistance and plant immune response.This provides a theoretical basis for studying the symbiotic relationship between soya and rhizobia.
