摘要
[目的]克隆获得凡纳滨对虾自噬相关基因Lv ATG6 cDNA全长,揭示其组织表达特征及其在白斑综合症病毒(WSSV)感染后的表达变化。[方法]利用PCR方法克隆Lv ATG6,半定量PCR方法检测Lv ATG6在对虾10种组织中的表达情况及WSSV侵染后Lv ATG6在对虾鳃和肝胰脏2种组织中的基因表达变化。[结果]克隆获得Lv ATG6 cDNA全长1 275 bp,编码424个氨基酸,预测蛋白分子量大小为51.5 k D;Lv ATG6基因在凡纳滨对虾10种组织中均有表达,无组织表达特异性;Lv ATG6在病毒感染不同时间的对虾鳃和肝胰脏组织中均出现明显的表达变化。[结论]凡纳滨对虾Lv ATG6及参与的细胞自噬可能在WSSV病毒感染增殖过程中发挥了重要作用,该研究为深入揭示凡纳滨对虾细胞自噬与WSSV病毒作用关系奠定了基础。
[Objective]To clone a full length of autophagy related gene 6(LvATG6)and reveal its tissue expression characteristics in Litopenaeus vannamei.To detect the changes of LvATG6 tissue expression after WSSV infection.[Method]LvATG6 of L.vannamei was cloned by PCR.The expression level of LvATG6 in 10 tissues of L.vannamei and gene expression changes of LvATG6 in gills and hepatopancreas after WSSV infection were detected by semiquantitative PCR.[Result]The full length of LvATG6 cDNA was obtaind,including 1 275 bp which encoded 424 amino acids.The molecular weight of predicted protein was 51.5 kD.LvATG6 expressed in 10 tissues of L.vannamei,without tissue expression specificity.LvATG6 showed significant expression changes in the gills and hepatopancreas of the shrimp after WSSV infection.[Conclusion]LvATG6 and autophagy of this gene involved in may play an important role in the proliferation of WSSV infection.This study provides a basis for revealing the relationship between autophagy and WSSV virus in L.vannamei.
