过氧化物酶V基因沉默HepG2肝癌细胞系的构建及鉴定 被引量:3

摘要 过氧化物酶V(Peroxiredoxin V,Prx V)是过氧化物酶家族(Peroxiredoxins)中的一员,具有清除细胞内活性氧(Reactive Oxygen Species,ROS)从而抑制由于氧化应激导致的细胞凋亡的作用。该研究通过慢病毒载体构建Prx V基因沉默的HepG2肝癌稳定细胞系。采用荧光照相及蛋白免疫印迹法对其结果进行鉴定,为研究Prx V的功能提供良好的基础。结果表明:成功构建Prx V基因沉默的HepG2肝癌细胞系。 Peroxiredoxin V(Prx V)was a thioredoxin peroxidase involved in peroxiredoxins family and it was also known as reactive oxygen species(ROS)scavenger which played a role in protecting the oxidative stress induced cell apoptosis.The Prx V knockdown HepG2 cell line was established throughlentivirus vectors and was analyzed the protein levels by western blot.The results showed that throughthe lentivirus vector treatments the Prx V knockdown HepG2 cell line was successfully established.
