摘要
为了探究朊病毒感染中钙调蛋白信号通路的变化情况,对羊瘙痒因子263K感染仓鼠脑组织中钙调蛋白的表达变化、分布特点及下游相关激酶及调控蛋白的表达变化情况进行了检测。结果显示在羊瘙痒因子263K感染仓鼠终末期脑组织中钙调蛋白的表达水平较正常仓鼠明显上调。钙调蛋白下游相关激酶及它们的磷酸化形式均明显下调,同时Ca MKs的下游调控蛋白——CREB也呈下调趋势,而靶蛋白——BDNF的表达则明显受到抑制。研究提示在朊病毒感染中很可能存在某种原因影响了钙调蛋白的功能。
To study the alterations of calmodulin signal pathway during prion infection,the expression changes and distribution pattern of calmodulin,expression levels of calmodulin related kinase,as well as downstream regulatory and target protein in the brain tissues of hamsters infected by scrapie factor 263K were analyzed.High level of calmodulin in the brain tissues of hamsters infected by 263K was observed by Western blot.Subsequent results showed calmodulin-related kinases and their phosphorylated forms were remarkable down regulated,meanwhile,the expression level of the downstream protein of calmodulin kinases,CREB was extremely decreased,while the expression of target protein BDNF was inhibited obviously.The study indicated that there might exist some reason to affect the function of calmodulin during prion infection.
