摘要
目的:研究维泰醇(alternol)对人黑素瘤细胞A375的增殖及黑素生成的影响。方法:不同浓度(0、0.3、0.6、1.5、3.0和4.0μg/ml)的维泰醇作用于A375细胞24h或48h,以MTT法观察药物对细胞增殖的抑制率。用台盼蓝拒染法测定药物作用后细胞存活率的变化,用Giemsa染色法在普通光镜下观察维泰醇对A375细胞形态的影响,用氢氧化钠裂解法测定维泰醇对A375细胞黑素生成的影响,用多巴氧化法测定其对酪氨酸酶活性的影响。结果:经维泰醇作用24h或48h后,细胞生长受到明显抑制,并呈时间和浓度依赖性。维泰醇作用后,在普通光镜下观察Giemsa染色结果,发现A375细胞出现很多树突状分支,且随着药物浓度的增大,分支越来越明显。维泰醇作用于A375细胞后,细胞内和细胞外的黑素含量明显增加,酪氨酸酶活性也呈现上升趋势。结论:维泰醇可抑制人黑素瘤A375细胞增殖,药物作用于细胞24h后,对细胞存活率有明显抑制作用。维泰醇能浓度依赖性地增加细胞中黑素生成和酪氨酸酶活性。
Objective:To study the effects of alternol on the proliferation and melanin synthesis of human A 375 melanoma cells.Methods:A375 melanoma cells were incubated with different concentrations(0,0.3,0.6,1.5,3.0 and 4.0^g/ml)of alter-nol for 24 and 48 h.Subsequently,the inhibition rate of cell proliferation was detected by tetrazolium salt colorimetric assay(MTT).Survival rate of A375 cells treated with alternol was determined by trypan blue exclusion staining test.The morpholog-ical changes of A375 cells were detected by Giemsa staining.Sodium hydroxy alkaline lysis method and dopa oxidation method were used to measure the content of melanin and activity of tyrosinase,respectively.Results:Alternol inhibited the proliferation of A375 cells significantly after incubation for 24 or 48 h in a time-dependent and concentration-dependent manner.After treat-ment with alternol,obvious morphological changes of A375 cells were observed,i.e.dentric projections.Tyrosinase activities and melanin content were increased obviously with the increase of alternol concentration.Conclusion:Alternol could surpress the cell proliferation,change cellular morphology,increase melanin level and tyrosinase activity of A 375 cells in a concentration-dependent manner.
