黄芪多糖和黄芪甲苷诱导RIMVECs分泌IFN-γ的研究 被引量:4

摘要 【目的】探明黄芪甲苷和黄芪多糖能否诱导RIMVECs分泌IFN-γ,为黄芪及有效成分的临床应用提供理论和参考依据。【方法】以体外培养的RIMVECs为模型,用ELISA法检测不同浓度的黄芪甲苷和黄芪多糖在不同时间诱导RIMVECs分泌IFN-γ的表达量。【结果】25μg/mL和50μg/mL黄芪甲苷能诱导RIMVECs表达IFN-γ,且随着时间延长表达逐渐升高,24h时达到峰值,具有明显的富集效应;25μg/mL和50μg/mL黄芪多糖也能诱导RIMVECs表达IFN-γ,随着时间延长RIMVECs细胞上清液中IFN-γ表达量逐渐增多,25μg/mL组IFN-γ表达量24h达到峰值;50μg/mL组IFN-γ表达量12h达到峰值,随后表达量降低。【结论】结果提示,黄芪甲苷和黄芪多糖的免疫调节作用可能是通过诱导RIMVECs分泌IFN-γ来实现,为黄芪的临床应用提供了试验依据。 【Objective】To provide a theoretical basis and reference for the clinical use of the effective components of Astragalus,the present study detected the Astragalus Polysaccharide and Astragaloside effect on IFN-γsecretion of RIMVECs.【Methods】Astragalus saponins and Astragalus polysaccharide are used to induce the IFN-γsecretion of RIMVECs,The IFN-γare detected with ELISA method.【Results】The expression IFN-γin RIMVECs gradually increased over time in 25 ug/mL and 50 ug/mL Astragaloside treated group,it reached the peak after 24 h with obvious enrichment effect;the expression of IFN-y was also gradually increased over time in 25 ug/mL Astragalus polysaccharide treated group,it reached the highest expression level after 24 50 ug/mL Astragalus polysaccharide could also increase the IFN-γexpression over time with a peak after 12 h,then the expression level decreased.【Conclusion】The results suggested that the immunomodulatory effects of Astragaloside and Astragalus polysaccharide might induce the IFN-γsecretion of RIMVECs,which provided experimental basis for the clinical application of Astragalus membranaceus.
