摘要
通过响应面法对一株产琼胶酶菌株的发酵工艺进行了优化,得到最佳发酵培养基组分为:酵母粉0.42%、琼脂粉0.37%,NaCl 3.3%,MgSO_4 0.5%,KCl 0.1%,CaCl_2 0.02%,K_2HPO_4 0.01%,FeSO_4 0.002%;最佳发酵条件为:发酵温度31℃,装液量为250mL三角瓶装100mL培养基,接种量4%,pH 7.8。经验证,最优条件下的酶活力达到36.84±1.9U/mL,较优化前酶活力增加了45.61%。
The fermentation technology of a strain producing agarose was optimized by several methods,results showed that the optimal culture medium consisted of 0.42%yeast extract,0.37%agar,3.3%NaCl,0.5%MgSO 4,0.1%KCl,0.02%CaCl 2,0.01%K 2HPO 4,0.002%FeSO 4 and the optimum enzyme producing conditions of the strains were:pH 7.8 at 31℃,100 mL culture medium in 250 mL flask,cultivating for 24 hours.Under the above conditions,the activity reached 36.84±1.9 U/mL,which was 45.61%higher than before.
