共培养体系中猪骨骼肌卫星细胞对肌内前体脂肪细胞脂质沉积的影响 被引量:1

Effects of Porcine Skeletal Muscle Satellite Cells on Lipid Deposition in Intramuscular Preadipocytes in Co-Culture System

摘要 本试验旨在研究共培养体系中猪骨骼肌卫星细胞对肌内前体脂肪细胞脂质沉积的影响。试验分离培养了猪骨骼肌卫星细胞和肌内前体脂肪细胞,2种细胞分离培养鉴定后,接种于Transwell细胞小室共培养板进行共培养,待细胞密度达到90%以上分别进行诱导分化。骨骼肌卫星细胞诱导分化8 d后,检测肌内前体脂肪细胞分化水平、分化标志基因的表达以及脂质代谢关键酶的表达。结果显示:与肌内前体脂肪细胞单独培养相比,共培养体系内肌内前体脂肪细胞的脂滴数量和脂滴面积极显著减少(P<0.01),肌内前体脂肪细胞增殖分化转录因子过氧化物酶体增殖物激活受体γ和CCAAT增强子结合蛋白的基因和蛋白表达水平极显著下降(P<0.01),肌内前体脂肪细胞的乙酰辅酶A羧化酶、脂肪酸合成酶的基因和蛋白表达水平极显著下降(P<0.01)。由此可见,共培养体系中猪骨骼肌卫星细胞对肌内前体脂肪细胞脂质沉积具有抑制作用。 This experiment was conducted to study the effects of porcine skeletal muscle satellite cells on lipid deposition in intramuscular preadipocytes in co-culture system.Porcine skeletal muscle satellite cells and intramuscular preadipocytes were isolated and cultured,after the two cells were isolated and identified,they were co-cultured in Transwell cell co-culture plates,and the cells were inducted differentiation with the density above 90%.The intramuscular preadipocytes differentiation level,the expression of differentiation marker gene and the key enzymes in lipid metabolism were detected after differentiation of skeletal muscle satellite cells for 8 days.The results showed that compared with intramuscular preadipocytes alone culture,in the co-culture system,the lipid droplet number and lipid droplet area in intramuscular preadipocytes were significantly decreased(P<0.01),the gene and protein expressions of peroxisome proliferators-activated receptorγand CCAAT enhancer binding proteinαin intramuscular preadipocytes were significantly decreased(P<0.01),the gene and protein expressions of acetyl CoA carboxylase and fatty acid synthase in intramuscular preadipocytes were significantly decreased(P<0.01).It is concluded that porcine skeletal muscle satellite cells have inhibitory effect on lipid deposition in intramuscular preadipocytes in the co-culture system.
