MicroRNA-451和microRNA-503对羟基脲诱导的K562细胞γ珠蛋白基因表达的调控作用

摘要 目的:研究羟基脲对K562细胞γ珠蛋白基因表达的影响及其中可能涉及的microRNA(miR)调控作用,以探讨药物治疗β地中海贫血的新思路。方法:用不同浓度(0μmol/L、100μmol/L、200μmol/L、300μmol/L、400μmol/L)的羟基脲作用于K562细胞24h、48h、72h、96h,采用实时荧光定量PCR(qPCR)检测γ珠蛋白及相关miR基因相对表达水平。结果:K562细胞γ珠蛋白基因表达显著增高(P<0.05),miR-451a和miR-503-3p基因表达明显升高(P<0.01)。结论:羟基脲能够诱导K562细胞γ珠蛋白基因高表达,其对γ珠蛋白基因的诱导作用可能通过促进miR-451a和miR-503-3p的表达来进行调控。 Objective:To study the effects of hydroxyurea onγglobin gene expression in K562 cells and the regulation of microRNA that may be involved.Methods:K562 cells were treated with different concentrations of hydroxyurea(0μmol/L,100μmol/L,200μmol/L,300μmol/L,and 400μmol/L)for 24 h,48 h,72 h and 96 h.Real-time fluorescent quantitative PCR(qPCR)was used to detect the expression ofγglobin and relevant microRNA.Results:The expression ofγglobin gene,miR-451a and miR-503-3p in K562 cells were significant increased in the hydroxyurea groups,compared with the control group(P<0.05 or P<0.01).Conclusion:Hydroxyurea could up-regulateγglobin gene expression in K562 cells,probably though the modulation of miR-451a and miR-503-3p expressions.
