摘要
目的:研究多西环素对烟曲霉菌性角膜炎中炎症细胞及单核细胞趋化蛋白-1(MCP-1)表达的影响。方法 :取健康SD大鼠64只,随机分为对照组和多西环素组,每组32只。两组均建立大鼠烟曲霉菌性角膜炎模型,多西环素组给予0.3g/L多西环素眼液点眼,对照组给予眼液溶媒点眼。分别于第3、第5、第7、第10天观察角膜炎症指数,分别采用荧光定量PCR(qPCR)法和酶联免疫吸附试验(ELISA)法检测MCP-1基因和蛋白的表达水平。结果:多西环素组第3、第5、第7天的炎症指数均低于对照组(P<0.05),各时间点炎症细胞计数均明显少于对照组(P<0.05);与对照组比较,多西环素组第3、第5、第7天MCP-1表达量减少(P<0.05)。结论:多西环素作用于大鼠烟曲霉菌性角膜炎可减少炎症细胞的浸润,并抑制炎症因子MCP-1的表达。
Objective:To investigate the effect of doxycycline on the inflammatory cells and the expression of monocyte chemoattractant protein-1(MCP-1)in rats with Aspergillus fumigatus keratitis(AFK).Methods:Sixty-four SD rats were randomly divided into a control group and a doxycycline group,with 32 rats in each group.All rats were established the AFK model.Rats in the doxycycline group were treated with 0.3 g/L doxycycline eye-drops,and those in the control group received menstruum eye-drops.Corneal inflammatory index was recorded on the 3rd,5th,7th,and 10th day.The mRNA and protein levels of MCP-1 were detected by fluorescence quantitative PCR(qPCR)and enzyme-linked immunosorbent assay(ELISA),respectively.Results:Compared with the control group,the inflammatory index was decreased in the doxycycline group on the 3rd,5th,and 7th day,and the inflammatory cell count was reduced at any time points(P<0.05).The expression of MCP-1 mRNA and protein was down-regulated on the 3rd,5th and 7th day after doxycycline treatment(P<0.05).Conclusion:Doxycycline could decrease the infiltration of inflammatory cells in rats with AFK,the mechanism might be related to the inhibition of MCP-1 expression.
