电针对糖尿病周围神经病变大鼠脊髓脂氧合酶的影响 被引量:4

Effects of Electroacupuncture Therapy on Expression of Spinal Lipoxygenase in Rats of Diabetic Peripheral Neuropathy

摘要 目的观察电针对糖尿病周围神经病变(DPN)大鼠脊髓脂氧合酶(LOX)的影响,探讨其干预DPN的作用机制。方法 30只实验大鼠随机分为正常组、模型组、电针组、脂氧素组、黄芩素组,每组6只,模型组大鼠采用单次腹腔注射链脲佐菌素造模,尾静脉采血检测血糖,筛选DPN动物模型,不干预。电针组大鼠选取"肾俞""足三里"电针2周;脂氧素组大鼠脊髓鞘内注射脂氧素3次;黄芩素组大鼠脊髓鞘内注射黄芩素3次。电子佛莱毛检测大鼠机械痛行为学,RT-PCR检测大鼠LOX基因水平,ELISA检测大鼠坐骨神经髓鞘碱性蛋白(MBP)表达。结果与正常组比较,模型组大鼠机械痛阈值及坐骨神经MBP含量显著降低(P<0.01),大鼠脊髓5-LOX、12-LOX、15-LOX mRNA水平显著升高(P<0.01);与模型组比较,电针组、脂氧素组和黄芩素组大鼠机械痛阈值和坐骨神经MBP含量显著升高,脂氧素组大鼠脊髓5-LOX mRNA表达显著降低(P<0.01),电针组和黄芩素组大鼠脊髓5-LOX、12-LOX、15-LOX mRNA表达显著降低(P<0.01)。结论电针能减轻DPN大鼠痛敏反应,修复坐骨神经损伤,其机制可能与抑制脊髓LOX活性有关。 Objective To observe the effects of electroacupuncture therapy on the expression of spinal lipoxygenase(LOX)in rats of diabetic peripheral neuropathy(DPN);To discuss its mechanism in intervening DPN.Methods Thirty experimental rats were randomly divided into five groups:normal,DPN,electroacupuncture,LXA4 and Baicalein,6 rats in each group.The model group was injected intraperitoneally with streptozotocin(STZ),and the animal model of DPN was screened by blood sampling of tail vein with blood glucose,without intervention.Electroacupuncture was applied to bilateral“Shenshu”and“Zusanli”acupoints in electroacupuncture group for two weeks.The LXA4 and Baicalein groups were successfully intrathecal injected 3 times with LXA4 and Baicalein in the DPN rats.The paw withdrawal threshold was measured with electronic Frye fibers.The mRNA levels of LOX in the L4~5 spinal cord of DPN rats were detected with real-time PCR analysis.The protein concentration of sciatic nerve MBP was detected with ELISA analysis.Results Compared with the normal group,the mechanical pain threshold and sciatic nerve MBP content in the model group significantly decreased(P<0.01),and the levels of 5-LOX,12-LOX and 15-LOX mRNA in the spinal cord of the model group significantly increased(P<0.01).Compared with model group,the mechanical pain threshold and MBP content of sciatic nerve in electroacupuncture group,LXA4 group and Baicalein group significantly increased,and the 5-LOX mRNA level in spinal cord of LXA4 group significantly decreased(P<0.01).The expressions of 5-LOX,12-LOX and 15-LOX mRNA in electroacupuncture group and Baicalein group significantly decreased(P<0.01).Conclusion Electroacupuncture therapy may reduce the pain response and restore the impairment of sciatic nerve in DPN rats through inhibiting the gene expression of LOXs in spinal cord.
