二氢杨梅素对绒癌JAR细胞MMP-2、PI3K和Akt表达的影响 被引量:4

摘要 目的:研究二氢杨梅素(dihydromyricetin,DMY)对绒癌JAR细胞基质金属蛋白酶2(matrix metalloproteinase2,MMP-2)mRNA、细胞磷脂酰肌醇3-激酶(phosphatidylinositol3-kinase,PI3K)mRNA和蛋白激酶B(protein kinase B,Akt)mRNA表达的影响。方法:采用实时荧光定量PCR法检测浓度为60μg/ml的二氢杨梅素作用36h,绒癌JAR细胞MMP-2 mRNA、PI3K mRNA、Akt mRNA表达的变化,并分析JAR细胞MMP-2 mRNA表达水平与PI3K mRNA、Akt mRNA表达水平的相关性。结果:与对照组相比,浓度为60μg/ml的二氢杨梅素作用36h,JAR细胞MMP-2 mRNA、PI3K mRNA、Akt mRNA的表达水平明显降低(P<0.05);并且,JAR细胞MMP-2 mRNA表达水平与PI3K mRNA、Akt mRNA表达水平呈正相关关系(r=0.957、0.953,P<0.05)。结论:二氢杨梅素可能通过下调绒癌JAR细胞MMP-2基因的表达影响JAR细胞的侵袭和转移,其机制可能与影响PI3K/Akt信号通路有关。 Objective:To investigate the effects of dihydromyricetin on matrix metalloproteinase 2(MMP-2),phosphatidylinositol3-kinase(PI3K)and protein kinase B(Akt)mRNA expression in choriocarcinoma JAR cells.Methods:Real time fluorescence quantitative PCR was used to detect the MMP-2,PI3K and Akt mRNAexpression in JAR cells after the JAR cells were treated with dihydromyricetin(60μg/ml)for 36h;as well as the correlations between MMP-2 mRNA and PI3K,Akt mRNA in JAR cells were analyzed.Results:After the JAR cells were treated with dihydromyricetin(60μg/ml)for 36h,the MMP-2,PI3K and Akt mRNA expression in JAR cells significantly decreased compared with control group(P<0.05).Moreover,the MMP-2 mRNAexpression in JAR cells was positively correlated with PI3K and Akt mRNA expression(r=0.957,0.953,P<0.05).Conclusions:Dihydromyricetin may in fluence the invasion and metastasis of JAR cells by down regulating the MMP-2 gene expression in JAR cells,the mechanisms maybe related to the effects of dihydromyricetin on PI3K/Akt signaling pathway.
