晚清书画裱件一株霉菌的鉴定及培养条件优化 被引量:3

摘要 对晚清书画裱件菌斑菌株复苏分离鉴定及培养条件优化.以改良马铃薯葡萄糖琼脂培养基、萨氏培养基和查氏培养基作为复苏培养基.采用三点植入法和插片法对菌株形态学观察,通过PCR扩增、测序、构建系统发育树对菌株进行分子生物学鉴定.结果成功复苏一株霉菌,菌株鉴定为篮状菌属,最佳培养条件为改良后PDA培养基,培养温度30℃,pH 6.0,震荡频率130 r/min. We revived,isolated,identified,and optimized the recovery of the mold on the late Qing Dynasty painting and calligraphy.The modified potato dextrose agar culture and SA culture medium serve as the reviving culture medium.The morphological observation was carried out by three point method and insert method;the molecular biological identification was carried out by PCR amplification,sequencing and the establishment of the phylogenetic tree.A strain of fungus was successfully recovered in the experiment.The optimum cultivation culture is the PDA culture medium,temperature 30℃,oscillation frequency 130 r/min,pH6.0.
