摘要
马属动物是嗜吞噬细胞无浆体的重要天然宿主,旨在弄清新疆昭苏县马感染嗜吞噬细胞无浆体情况和病原特征。对采集的100份马血清进行间接免疫荧光(IFA)检测嗜吞噬细胞无浆体抗体,对阳性样品相对应的DNA样品通过PCR扩增、16S rRNA基因测序、序列比对、完成了分子分型分析。间接免疫荧光法(IFA)检出率为8%(8/100),通过对阳性样品的基因组进行16S rRNA扩增,序列分析进一步确定马存在嗜吞噬细胞无浆体感染情况。分子分型发现存在2种嗜吞噬细胞无浆体基因型,ZS23和ZS40这2种基因型在我国及周边其他国家均有分布,研究结果将为该地区的嗜吞噬细胞无浆体感染情况的调查和防治提供依据。
Horse is an important natural host of Anaplasma phagocytophilum.The aim of this study was to investigate A.phagocytophilum infection and characterize the pathogen in horses in Zhaosu county of Xinjiang province.In this study,an indirect fluorescent antibody assay(IFA)was used to detect A.phagocytophilum antibodies,and the corresponding DNA samples of the antibody positive samples were subjected to PCR detection and sequencing on the basis of 16S rRNA gene(641 bp).The obtained sequences were then used for genetic identification of the pathogen.The serological positive rate of A.phagocytophilum infection was 8%(8/100).The PCR detection further confirmed the infection of A.phagocytophilum in horses in the investigated regions.Genetic identification found two epidemic strains,which were highly similar to the ones previously reported in China or in its neighboring countries.These data will be helpful for epidemiological investigation or making control measures of the A.phagocytophilum infection in the surveyed region.
