摘要
探讨炙甘草汤对慢性疲劳综合征(CFS)大鼠行为学和血清IL-1β、IL-2、IL-6、IL-12表达水平的影响。通过慢性束缚法加冷水游泳法建立慢性疲劳综合征大鼠模型(4周),将30只造模成功大鼠随机分为模型组(2mL/kg)和炙甘草汤组(12g/kg),15只未造模大鼠为对照组(2mL/kg)。共灌胃2周。观察各组大鼠旷场试验、力竭游泳试验、体质量的变化,并检测各组大鼠血清IL-1β、IL-2、IL-6、IL-12的水平。结果表明,与对照组比较,模型组力竭游泳时间、穿格次数、直立次数、体质量明显减少(P<0.05),血清IL-1β、IL-6水平显著升高(P<0.05),IL-2、IL-12的水平显著降低(P<0.05);与模型组比较,炙甘草汤组力竭游泳时间、穿格次数、直立次数、体质量显著升高(P<0.05),血清IL-1β、IL-6水平显著降低(P<0.05),IL-2、IL-12的水平显著升高(P<0.05)。说明炙甘草汤可能通过抑制血清IL-1β、IL-6的表达,促进IL-2、IL-12的表达,调节免疫功能,减轻疲劳症状,发挥治疗慢性疲劳综合征的作用。
This study aimed to study the effects of prepared licorice decoction on behavioristics and serum IL-1β,IL-2,IL-6,IL-12 in rats with chronic fatigue syndrome(CFS).CFS rat model was induced by chronic restraint and swimming in cold water for 4 weeks.Then 30 CFS rats were randomly divided into the model group(2 mL/kg)and the prepared licorice decoction group(12 g/kg),and 15 rats were selected as the control group(2 mL/kg).Behavioristics and the levels of serum IL-1β,IL-2,IL-6,IL-12 were measured before and after drug intervention for 2 weeks.The results showed that,compared with the rats of the control group,the model group manifested a series of abnormal variations in Open-field test,exhausting swimming test and weight,the levels of serum IL-1β,IL-6 increased significantly(P<0.05),while IL-2,IL-12 decreased(P<0.05).Compared with the rats of the model group,the times of crossing lattice in Open-field test,the times of flounces,and the time of swimming till exhaustion and weight increased significantly(P<0.05),the levels of serum IL-1β,IL-6 decreased significantly(P<0.05),while IL-2,IL-12 increased(P<0.05).Prepared licorice decoction could treat CFS by reducing the levels of serum IL-1β,IL-6 and upregulating IL-2,IL-12 and improving fatigue symptoms.
