摘要
通过观察芒果苷(MGG)对人肝癌HepG2细胞生长、迁移能力、侵袭能力、细胞骨架及细胞周期等的影响,探讨MGG对肝癌细胞侵袭转移的作用及机制.以肝癌HepG2细胞为靶细胞,采用MTT法检测HepG2细胞的活力,采用划痕实验、侵袭实验测定HepG2细胞的体外迁移、侵袭能力;采用细胞骨架染色法观察细胞骨架;采用细胞流式仪测定细胞周期;采用Western blot检测HepG2细胞Cyclin D1和MMP9活性及蛋白表达.实验结果表明,MGG能抑制HepG2细胞的生长、迁移和侵袭,并呈现剂量依赖性;MGG可减少HepG2细胞微丝数量,降低细胞骨架的面积,MGG能减少S、G2/M期的细胞,抑制细胞分裂,并能抑制HepG2细胞中Cyclin D1和MMP9蛋白表达.因此,MGG能抑制肝癌HepG2细胞的生长、迁移及侵袭,具有潜在的抗肿瘤转移作用,其机制可能与MGG能影响细胞周期进程、细胞骨架重建及细胞外基质水解相关.
The mechanism of mangiferin in the inhibition of cancer cell invasion and metastasis is explored by investigating the effect of MGG on cell migration,cell invasion and cytoskeleton of HepG2 cells.Using HepG2 cells as target cells,Cell viability was measured by 3-(4,5-dimethyl-2-thia.zolyl)-2,5-diphenyltetrazolium bromide(MTT).Cell invasion and migration of different concentrations were measured by Transwell assay and Scratch assay.The effect of MGG on cyto.skeleton of HepG2 cells was detected by FITC Phalloidin.The distribution of cell cycle was ana.lyze by Flow cytometry.The expression of HepG2 cells CyclinD1 and MMP9 were detected by western blot.MGG significantly inhibited the proliferation,migration and invasion of HepG2 cells,and showed a dose-dependent manner.Flow cytometry analysis showed that different concentrations of pekinenal could make the cell block in S phase.In consequence,MGG can in.hibit the growth,migration and invasion of HepG2 cells,and it has the potential to resist tumor me.tastasis,and the mechanism may be associated with MGG significantly decreasing the number and area of cytoskeleton,and inhibiting the proliferation and the express of CyclinD1 and MMP9.
