摘要
通过生物大数据和检测工具,对烟草半胱氨酸蛋白酶的序列特征、亚细胞定位、二级结构和三级结构进行分析。结果显示,烟草半胱氨酸蛋白酶均具有信号肽,且含有组织蛋白酶前肽抑制剂结构域和木瓜蛋白酶家族半胱氨酸蛋白酶;经初次亚细胞定位,NtCP-1,NtCP-2,NtCP-3和NtCP-4定位于细胞质,NtCP-5定位于液泡;二级结构中,NtCP-1,NtCP-2和NtCP-5的α-螺旋比例最大,而NtCP-3和NtCP-4以无规则卷曲比例最大;三级结构N-端聚集较为疏松,其二级结构以α-螺旋为主,C-端的二级结构以β-折叠为主。
Tobacco cysteine protease sequence characteristics,subcellular localization,secondary structure and tertiary structure were analyzed with large biological data and detection tools.The results showed that tobacco cysteine protease had a signal peptide,and contained a cathepsin propeptide inhibitor domain and a papain family cysteine protease.The initial subcellular localization,NtCP-1,NtCP-2,NtCP-3 and NtCP-4 were located in cytoplasm,and NtCP-5 was located in the vacuole.In the secondary structure,NtCP-1,NtCP-2 and NtCP-5 had the largest proportion of alpha helix,while NtCP-3 and NtCP-4 had the largest specific gravity in the random coil.The tertiary structure had looser N-terminal aggregation,its secondary structure was mainlyα-helix,and the C-terminal secondary structure was mainlyβ-sheet.
