乳腺癌侵袭转移中Artemin作用的探讨 被引量:1

摘要 目的:探讨Artemin对乳腺癌侵袭转移的作用与意义。方法:采用免疫组织化学方法对乳腺癌组织切片进行Artemin表达分析。以乳腺癌MDA-MB-231细胞为研究对象,通过si RNA技术降低内源性ARTN的水平获得siATRN-MDA231稳定克隆细胞,通过Western blot技术验证siATRN-MDA231稳定克隆细胞ARTN表达水平变化。通过划痕和趋化运动实验观察细胞迁移和定向运动能力的变化。通过粘附和侵袭实验观察siATRN-MDA231细胞粘附功能以及侵袭能力变化。结果:乳腺癌组织中Artemin表达水平与肿瘤大小无关,但与淋巴结转移关系密切。siATRN-MDA231细胞Artemin的蛋白表达量明显比对照组降低,通过划痕和趋化实验显示肿瘤细胞的运动能力显著降低。细胞的粘附功能和侵袭水平也有不同程度减低。结论:Artemin参与EGF诱导的乳腺癌细胞的运动和侵袭。
