支气管哮喘患者与健康对照转录因子T-bet、GATA-3表达的比较 被引量:1

摘要 目的比较哮喘患者及健康对照外周血CD4^+T细胞转录因子T-bet、GATA-3表达水平。方法选取新疆医科大学第六附属医院门诊及住院哮喘患者100例;同期健康志愿者100例;获取的外周血标本中的CD4+T淋巴细胞,是采用免疫磁珠法分离提取获得;而T-bet、GATA-3转录因子m RNA表达水平,其检测用SYBR Green I实时荧光定量法。结果 T-bet基因m RNA表达含量在哮喘组中为(6.90±2.25),对照组中为(6.74±2.16)。与对照组相比,哮喘组表达含量较高,但差异无统计学意义(P>0.05)。GATA-3基因m RNA在哮喘中相对表达量为(4.22±2.03),健康对照组中相对表达量(3.15±2.22)。与对照组相比,哮喘组GATA-3m RNA明显较高,两组差异具有统计学意义(P<0.05)。结论支气管哮喘组Th2细胞转录因子GATA-3m RNA高表达,转录因子T-bet/GATA-3 m RNA的平衡破坏,进一步影响支气管哮喘的发生发展。 Objective To compare the expression levels of CD4+T cell transcription factor T-bet and GATA-3 were measured in patients with asthma and healthy controls.Methods 100 cases of outpatient and inpatient asthma in The Sixth Affiliated Hospital of Xinjiang Medical University and 100 healthy volunteers in the same period were selected.Peripheral blood samples of these patients were obtained,isolated peripheral blood mononuclear cells(PBMC)were isolated,and CD4+T lymphocytes were isolated by immune magnetic beads method.The expression level of T-bet and GATA-3 gene was detected by SYBR Green I real-time quantitative PCR.Results T-bet mRNA expression level in asthma group(6.90±2.25),and in the control group(6.74±2.16).The T-bet mRNA expression level of asthma group was higher than that in the control group,but the difference was not statistically significant(P>0.05).GATA-3 mRNA expression level in asthma group(4.22±2.03),and in the control group(3.15±2.22).The GATA-3 mRNA expression level of asthma group was higher than that in the control group,and the difference between the two groups was statistically significant(P<0.05).Conclusion The high expression of Th2 cell related transcription factor GATA-3 in peripheral blood of asthmatic patients,and the imbalance expression of transcription factor T-bet and GATA-3 in asthma patients showed that the imbalance expression affected the development of asthma.
